Flurbiprofen (INN, also known as Ansaid, Froben, Flurizan, or L-790,330) is a non-steroidal anti-inflammatory drug (NSAID) of the propionic acid derivative class. It functions as a potent, non-selective cyclooxygenase (COX) inhibitor with IC50 values of 0.1 µM and 0.4 µM for human COX-1 and COX-2, respectively. It exhibits anti-inflammatory, analgesic, antipyretic, neuroprotective, anti-metastatic, and anticancer chemotherapeutic activities. In vivo, flurbiprofen suppresses cerebral ischemia/reperfusion injury by decreasing infarct volume, apoptosis, and neurological deficits. In cellular and animal models of gastric cancer, it downregulates cyclin D1 and CDK4 expression, upregulates p75 expression, and decreases cellular proliferation, metastasis, and tumor size. Used as a pharmaceutical intermediate and active pharmaceutical ingredient (API) for the development and quality control of oral tablets, topical patches, ophthalmic solutions, and transdermal delivery systems. Approved for clinical use in the treatment of rheumatoid arthritis, osteoarthritis, ankylosing spondylitis, and mild to moderate pain.
