Nolatrexed (AG-337) is a non-classical thymidylate synthase (TS) inhibitor belonging to the non-classical antifolate class. It binds to the folate cofactor site of TS, competitively inhibiting TS activity (Ki = 11 nM), thereby blocking dTMP synthesis and inhibiting DNA synthesis and cell proliferation. It was developed primarily for hepatocellular carcinoma (HCC) and studied in head and neck cancers and other solid tumors. It is used as a key synthetic precursor/intermediate for Nolatrexed dihydrochloride and as a tool compound in TS resistance mechanism research and structure-based drug design.
